Semi-skirted PCR plates have a partial plastic border extending approximately half the height of a fully skirted plate, surrounding part of the well array. This partial skirt adds enough structural rigidity to support multichannel pipetting. It provides a surface for labelling, while keeping the overall plate height low enough for compatibility with most thermal cycler compression lids that a full skirt would obstruct. Semi-skirted plates represent the most common format in routine qPCR and genotyping labs, where fully automated robotic handling is not required. Semi-skirted is the default choice for most routine PCR and qPCR labs that mix manual pipetting with occasional automated steps.
Well Profile and Volume
Low-profile 0.1 mL semi-skirted plates are the standard for qPCR on fast-block instruments, including Bio-Rad CFX96, Applied Biosystems StepOnePlus, and Roche LightCycler 480. Standard-profile 0.2 mL wells work with conventional block cyclers and accommodate larger reaction volumes for cloning or high-concentration template workflows.
Colour: Clear vs White
Clear semi-skirted plates suit end-point PCR and top-read qPCR systems. White semi-skirted plates increase fluorescence signal in bottom-read qPCR optical configurations. Thermo Fisher offers white semi-skirted formats for this application.
Cut Corner for Orientation
Semi-skirted plates typically feature a notched or cut corner at the A12 position for plate orientation — a feature originating in sequencing plate formats and now standard across most qPCR-compatible semi-skirted plates. Confirm the cut corner position against your instrument's plate alignment specification.
Sealing
The partial skirt of semi-skirted plates supports adhesive film sealing and is compatible with most heat sealers. The skirt also reduces edge-well evaporation compared to non-skirted plates, improving Ct uniformity across plate positions.
Robotic Compatibility
Semi-skirted plates provide enough grip surface for some robotic arm designs and are listed as compatible with select liquid handlers; however, for demanding robotic workflows with gripper systems or conveyor belts, fully skirted ANSI/SBS-compliant plates remain the correct specification.
Semi-skirted 96-well plates are moulded from virgin polypropylene with uniformly thin walls (0.25–0.35 mm) for consistent thermal conductivity. The partial skirt height is approximately 50% of a full ANSI/SBS skirt height, placing the plate between non-skirted and fully skirted in both rigidity and cycler compatibility. All plates are independently certified DNase-free, RNase-free, human DNA-free, and PCR inhibitor-free per lot.
