What Are Single-Channel Pipettors?


A single-channel pipettor is the single most widely used liquid handling instrument in molecular biology, cell biology, and clinical research. It transfers precise volumes between 0.1 µL and 5,000 µL, one well or tube at a time, using an air-displacement piston mechanism. The piston creates a partial vacuum that aspirates liquid into a disposable tip; advancing the piston fully dispenses the liquid. A spring-loaded tip ejector releases the used tip without hand contact for contamination control. Manual models depend on finger force and offer simple, battery-free operation; electronic models add programmable aspiration/dispensing speeds, multi-dispense modes, and up to 5 custom pipetting programs.  A set of four manual single-channel pipettors (P10, P20, P200, P1000) covers 95% of molecular biology liquid handling; add electronic models for high-throughput repetitive work.

How to Choose a Single-Channel Pipettor


Volume Range: Build a Complete Bench Set
A standard four-pipettor bench set covers P10 (1–10 µL), P20 (2–20 µL), P200 (20–200 µL), and P1000 (100–1,000 µL). For sub-microliter work (sequencing, single-cell), add a P2 (0.2–2 µL). For large-volume dispensing (media prep, column loading), add a P5000 (1,000–5,000 µL). Avoid selecting a volume range that places your target dispense below 20% of the maximum — accuracy degrades at the low end of any range.

Manual vs Electronic
Manual single-channel pipettors (Eppendorf Research Plus, Gilson PIPETMAN Classic, Rainin Pipet-Lite XLS+) are the standard for routine lab work: field-calibratable, autoclavable, and requiring no power source. Electronic single-channel pipettors (Eppendorf Research Pro, Rainin Pipet-Lite XLS+ Electronic) add: programmable aspiration/dispense speed (5 speeds), multi-dispense (repeat pipetting) mode, mix mode, dilution mode, and backlight LCD volume display. Electronic models reduce hand fatigue in workflows exceeding 200–300 tip changes per day.

Tip Compatibility
Three tip systems cover the market: standard conical (universal fit — fits Eppendorf Research Plus, Gilson PIPETMAN, Rainin non-LTS, Thermo, Sartorius/BioHit); Rainin LTS cylindrical (requires LTS-specific tips — lower force insertion); Eppendorf epT.I.P.S. proprietary (tightest fit for Eppendorf instruments). Autoclavability and Chemical Resistance
Manual pipettors from Eppendorf, Gilson, and Rainin have fully autoclavable lower sections (121°C, 15 psi, 15 min). Electronic pipettors have autoclavable nose cones only — the electronic module cannot be autoclaved.

Calibration and Service
All single-channel pipettors are factory-calibrated per ISO 8655. Field recalibration tools are available for Eppendorf, Gilson, and Rainin to perform in-lab gravimetric verification without external service.

Eppendorf Research Plus and Gilson PIPETMAN Classic pipettors are fully autoclavable at 121°C, 15 psi, 15 minutes. Rainin Pipet-Lite XLS+ and electronic pipettors such as Eppendorf Research Pro have autoclavable nose cones and lower sections only — the electronic module cannot withstand steam sterilisation conditions.
Place your most common target volume at 50–80% of the pipettor's maximum. For example, for regular 160 µL dispenses, a 20–200 µL pipettor is more accurate than a 100–1,000 µL instrument set to 160 µL. Using a pipettor below 20% of its maximum increases systematic volume error significantly.
Eppendorf Research Plus accepts universal-fit conical tips and Eppendorf epT.I.P.S. proprietary tips. Gilson PIPETMAN Classic accepts universal-fit conical tips. Rainin Pipet-Lite XLS+ LTS requires LTS cylindrical tips (lower insertion force) and does not accept standard universal-fit tips. Confirm tip compatibility before purchasing a pipettor to avoid tip-instrument mismatch.
Per ISO 8655 Part 2, a well-calibrated 200 µL air-displacement pipettor should achieve ±0.8 µL systematic error and ≤0.3 µL random error (CV ≤0.15%) with distilled water at 20°C. The Eppendorf Research Plus 200 µL model meets this specification. Pipettors deviating from these limits require calibration adjustment.
ISO 8655 recommends calibration verification at a minimum every 6 months for routine research use; GLP and accredited lab protocols typically require quarterly calibration. Pipettors should also be verified immediately after any physical impact, liquid ingress event, or observation of tip drip between aspirations.
In-lab gravimetric calibration is possible with a model-specific calibration tool (from Eppendorf, Gilson, or Rainin), a calibrated analytical balance with 0.0001 g resolution, distilled water at 20°C, and a minimum of 10 dispenses per volume point. ISO 17025-accredited external calibration from the manufacturer produces a certified calibration certificate for compliance documentation.
