NEST BioFactory™ Systems are compact, single-use, multi-layer cell culture vessels manufactured from USP Class VI polystyrene for large-scale adherent cell culture. The BioFactory provides the growth kinetics and cell attachment characteristics of standard T-flasks, scaled up to growth areas from approximately 636 cm² (1-layer) to 25,440 cm² (40-layer) in a single vessel — making it the scale-up path from T-flask and roller bottle culture for vaccine production, monoclonal antibody manufacturing, viral vector production (lentivirus, AAV, adenovirus), and biopharmaceutical protein expression from adherent cell lines.

How to Choose the Right BioFactory™ Layer Count


Matching BioFactory Scale to Production Stage
The 1-layer BioFactory (≈636 cm²) is equivalent in surface area to approximately 8–9 standard T75 flasks — used for proof-of-concept scale-up studies transitioning from T-flask monolayer culture. The 2-layer BioFactory (≈1,272 cm²) is the standard for pilot-scale viral antigen production and early-phase process development. The 5-layer (≈3,180 cm²) and 10-layer (≈6,360 cm²) formats suit mid-scale production runs for research-grade vaccine lots, transfection-derived protein, and pre-clinical viral vector batches. The 40-layer (≈25,440 cm²) is an industrial-scale format for commercial vaccine, mAb, and biopharmaceutical production.

Closed Liquid Transfer and Contamination Reduction
NEST BioFactory™ systems feature a narrow-mouth cap port compatible with aseptic tubing for closed liquid transfer — media, wash buffers, trypsin, and harvest supernatant can be introduced and removed through the tube port without opening the vessel. This closed-system operation eliminates the open-pour steps that create contamination risk in multi-layer roller bottle workflows and enables BSL-2 containment of pathogen-bearing cultures without biosafety cabinet–dependent open-handling. Wide-mouth caps allow direct media pouring for non-closed workflows.

TC Treatment and Cell Compatibility
All NEST BioFactory™ layers are TC-treated by vacuum plasma using the same surface treatment process as NEST cell culture T-flasks. NEST has published VERO cell culture data showing that cell morphology, adherence, and growth kinetics in the BioFactory are equivalent to those in standard T-flasks — cell attachment density, proliferation rate, and morphology are not altered by the scale-up. This linear scalability is the primary engineering advantage of multi-layer vessels over microcarrier bioreactors and packed-bed systems for adherent cell–derived products requiring 2D culture validation.

Ultrasonic Welding and Particle Generation
The BioFactory layers are joined by ultrasonic welding without chemical adhesives or extrinsic ingredients. NEST's welding line design minimizes particle generation at the weld joints, an important quality attribute for pharmaceutical production contexts where subvisible particle counts in harvest media must meet compendial limits.

Automation and GMP Compatibility
NEST BioFactory™ systems are compatible with automated media handling equipment. Multiple BioFactory units can be connected in series through the tubing ports for further scale-out. The double-bagged sterile packaging format supports GMP cleanroom entry requirements. NEST provides validation documentation packages (sterilization data, extractable/leachable certificates, biocompatibility reports) supporting GMP authorization for industrial cell culture production.

NEST BioFactory™ Systems are manufactured from USP Class VI polystyrene in a Class 10,000 (ISO 7) dedicated cleanroom. All culture surfaces are TC-treated by vacuum plasma. Temperature range: approximately −86 °C to +64 °C. E-beam sterilized to SAL 10⁻⁶; non-pyrogenic; double-bagged sterile packaging. Vent caps contain 0.22 µm hydrophobic filters for gas exchange without contamination. VERO cell culture validation data available from NEST Scientific: seeding at 5 × 10⁴/mL in 2-layer BioFactory (400 mL) shows robust, even adherence and growth kinetics matching T-flask controls after 3 days. Layer configuration depends on production volume: the 2-layer BioFactory (1,272 cm²) suits pilot-scale viral antigen production and process development runs; the 5-layer (3,180 cm²) and 10-layer (6,360 cm²) are standard for research-grade and pre-clinical vaccine lot production; the 40-layer (25,440 cm²) is used for commercial-scale vaccine manufacturing. Always determine inoculation density per cm² from T-flask growth data and multiply by BioFactory surface area to calculate seeding volume.
NEST BioFactory™ systems have a narrow-mouth tubing cap port compatible with aseptic tubing for closed media addition, wash, trypsin addition, and harvest operations without opening the vessel. This eliminates open-pour contamination risk and enables BSL-2 containment of pathogen-bearing cultures without requiring all media changes to be performed inside a biosafety cabinet. Wide-mouth caps are also available for direct media pouring in open-handling workflows.
