How to Choose PETG Erlenmeyer Flasks


PETG vs. PC for Shake Flask Culture
PETG flasks are single-use and cannot be autoclaved (maximum temperature +60 °C). PC flasks can be autoclaved 5–10 times and have a broader temperature range (−135 °C to +135 °C). For high-throughput suspension culture operations processing many flasks per week, single-use PETG eliminates all washing, autoclave queuing, and glassware inspection steps — reducing hands-on time and contamination risk from reuse. For labs with lower throughput that prepare media in-house via autoclave, PC remains the cost-effective choice. PETG is BPA-free; PC contains BPA.

Cell Line Compatibility
PETG Erlenmeyer flasks are validated for CHO-S, HEK293-F, Expi293, Jurkat, K562, Sf9 (Spodoptera frugiperda), Hi5 (Trichoplusia ni), S. cerevisiae, P. pastoris, and E. coli suspension culture. The non-treated PETG surface prevents inadvertent cell adhesion and is compatible with all standard suspension culture media, including FreeStyle CHO, Expi293, Sf-900, and ESF 921 insect cell media. PETG is chemically resistant to DMSO, ethanol, isopropanol, and standard aqueous buffers.

Baffled vs. Flat Base for PETG Flasks
Baffled-base PETG Erlenmeyer flasks are the standard for oxygen-demanding suspension cultures: CHO, HEK293-F, bacterial, and insect cell lines. The baffles generate turbulent flow during orbital shaking, raising dissolved oxygen and reducing cell sedimentation. For suspension culture with 20–25% fill volume, vented caps at standard orbital speeds (100–150 RPM) provide adequate oxygen mass transfer to support mammalian cell densities up to approximately 5 × 10⁶ cells/mL. For cultures targeting higher densities or with very high specific oxygen uptake rates (sOUR), confirm dissolved oxygen by inline monitoring before scaling further in PETG flasks.

Storage of Unused Sterile Flasks
Store unused NEST Scientific PETG Erlenmeyer flasks at room temperature (15–30 °C) in the original sealed sterile bag, away from UV light. Shelf life is typically 3 years from the production date per the lot certificate. Inspect the bag seal before use; a compromised seal invalidates sterility. Remove from the bag inside a biosafety cabinet immediately before inoculation.

Standard orbital shaker speeds for mammalian suspension culture in PETG baffled Erlenmeyer flasks are 100–150 RPM with a 25–50 mm orbital throw. Bacterial and yeast cultures in baffled PETG flasks typically require 150–250 RPM for adequate oxygen transfer. Confirm shear tolerance for your specific mammalian cell line before exceeding 150 RPM — above this threshold, shear-sensitive lines may show reduced viability.
Baffled-base PETG flasks have internal angled baffles that disrupt circular rotational flow during orbital shaking, generating turbulence that increases dissolved oxygen transfer by up to 2× compared to flat-base formats at the same RPM. Flat-base PETG flasks produce gentler, lower-shear rotational mixing suited for media preparation, gentle suspension of shear-sensitive cells, and culture stages where turbulence is undesirable.
