Erlenmeyer Flasks Accessories

What are Erlenmeyer Flask Accessories?

Erlenmeyer flask accessories are the replacement caps, transfer cap assemblies, vent filters, and adapter fittings used alongside PC and PETG shake flasks to customize gas exchange, enable closed-system liquid transfer, and replace consumable caps between culture runs. Accessory selection determines whether a shake flask acts as a standard culture vessel, a sealed storage container, or a closed-system liquid transfer assembly in a cGMP workflow.

How to Choose Erlenmeyer Flask Accessories

Vented Caps: Standard vs. Oversized Membrane Standard vented Erlenmeyer caps have a 0.22 µm PTFE membrane in a small-diameter HDPE housing. NEST Scientific vented caps feature an oversized membrane area that increases the gas exchange surface, supporting higher oxygen mass transfer for dense suspension cultures at elevated shaker speeds. At 150 RPM with 20–25% fill, NEST oversized vented caps provide sufficient oxygen transfer for mammalian suspension cultures at densities up to approximately 5 × 10⁶ cells/mL. For cultures targeting densities above 10 × 10⁶ cells/mL in a standard shake flask format, a sparged bioreactor system is more appropriate. Plug Seal Caps: Autoclave vs. Non-Autoclave HDPE plug seal caps for NEST PC Erlenmeyer flasks are compatible with autoclave cycles at 121 °C when loosened 1–1.5 turns before autoclaving to prevent steam pressure buildup. Plug seal caps for NEST PETG flasks are compatible with static incubation and transport but not autoclaving (PETG maximum temperature: +60 °C). Confirm whether your accessory cap is rated for autoclave use before including it in a steam sterilization cycle. Transfer Cap Assemblies for Closed-System Culture NEST customized transfer cap assemblies combine an Erlenmeyer-compatible cap body with a bi-directional transfer port, TPE tubing, and a Luer lock or CPC connector — enabling aseptic inoculation, sampling, or media addition to a running shake flask culture without opening the flask. This closed-system capability is used in cGMP process development, GMP seed train management, and contamination-critical research workflows. Transfer caps are available in sizes matching NEST 125 mL, 250 mL, 500 mL, 1,000 mL, 2,000 mL, and 3,000 mL flask neck formats. Cap Sizing and Flask Compatibility Erlenmeyer flask caps are sized to the neck thread diameter of the flask. NEST Scientific PC and PETG Erlenmeyer flasks use proprietary screw-thread neck dimensions — confirm the specific cap catalog number matches your flask's neck size before ordering replacement caps. Do not mix cap brands across flask brands without verifying thread compatibility; mismatched caps create gas leaks at the vent membrane, contaminating cultures. Replacement Cap Frequency for Reusable PC Flasks For reusable PC flasks, replace the vented cap after each culture run to prevent membrane clogging from media splatter, condensation, and biofilm formation on the PTFE membrane. Reusing a contaminated or clogged vent membrane reduces gas exchange efficiency, creating a DO deficit that depresses cell growth without a visible contamination event. NEST vented caps for PC flasks are available in bulk packs for cost-efficient per-run replacement.

Specifications Context

What are Cell Culture Erlenmeyer Flasks?

Cell culture Erlenmeyer flasks (also called shake flasks or conical flasks) are single-use, sterile plastic vessels with a conical body, flat base, and narrow neck used for suspension cell culture on orbital shakers and incubator shakers, microbial culture, and media preparation. Unlike rectangular T-flasks designed for static monolayer culture, Erlenmeyer flasks are engineered for shaking: their wide conical base maximizes liquid surface area for oxygen transfer, while the narrow neck minimizes evaporation and contamination risk. Choose PC Erlenmeyer flasks for autoclavable reuse and high-temperature media preparation; choose PETG for single-use suspension culture and processes requiring BPA-free materials.

How to Choose Erlenmeyer Flasks for Cell Culture

PC vs. PETG Material PC (polycarbonate) Erlenmeyer flasks are autoclavable at 121 °C (typically 5–10 cycles), shatter-proof, crystal-clear, and rated from −135 °C to +135 °C. PC is ideal for labs that sterilize media or wash in-house. PC contains BPA. PETG Erlenmeyer flasks are single-use, BPA-free, rated from −80 °C to +60 °C, and cannot be autoclaved. They provide a contamination-free, cleaning-free alternative to glass and PC for high-throughput suspension culture operations. Flat Base vs. Baffled Base Flat-base Erlenmeyer flasks provide gentler swirling for slow-growth cell lines and media mixing. Baffled-base flasks have three or four angled internal baffles that interrupt the swirling flow, increasing turbulence, raising dissolved oxygen (DO) by up to 2× compared to flat-base flasks at the same orbital speed, and reducing cell sedimentation. Baffled flasks are the standard for CHO-S, HEK293-F, Sf9, microbial, and yeast cultures requiring high oxygen transfer rates. NEST Scientific offers baffled and flat-base versions in both PC and PETG across all core volumes. Vented vs. Plug Seal Cap Vented caps contain a 0.22 µm hydrophobic PTFE membrane that allows free O₂/CO₂ gas exchange while excluding microbial contamination. The NEST vented cap features an oversized vent area that improves gas exchange beyond standard-size membrane caps, supporting oxygen-demanding suspension cultures at higher cell densities. Plug seal caps create a fully sealed environment for media storage, static incubation, or transport outside the incubator. Always use vented caps for actively growing suspension cultures on an orbital shaker. Volume and Fill Ratio Erlenmeyer flasks should be filled to no more than 20–25% of the nominal flask volume for suspension shaker culture to maintain adequate headspace for oxygen transfer. A 500 mL flask running at 20% fill holds 100 mL of culture; a 1,000 mL flask holds 200 mL. Overfilling reduces oxygen transfer efficiency and creates foam that blocks the vented cap membrane. For media storage (no shaking), the fill volume can approach 80% of nominal. Shaker Compatibility NEST Scientific Erlenmeyer flasks (125 mL–3,000 mL) are compatible with standard laboratory orbital shakers, including those from Infors HT, New Brunswick Scientific (Innova), and Thermo Scientific MaxQ. Confirm flask neck O.D. and base diameter against your shaker's flask holder clips or clamp system before ordering large quantities.

Specifications Context

What are PC Erlenmeyer Flasks?

PC (polycarbonate) Erlenmeyer flasks are autoclavable, shatter-proof, optically clear shake flasks manufactured from USP Class VI medical-grade polycarbonate for suspension cell culture, microbial fermentation, and media preparation on orbital shakers. Polycarbonate provides a working temperature range of −135 °C to +135 °C and withstands 5–10 autoclave cycles at 121 °C — the defining advantage of PC over PETG for labs that steam-sterilize media or require reusable, re-sterilizable shake flasks. PC Erlenmeyer flasks are the correct choice when autoclavability, reuse, or processing at elevated temperature (>60 °C) is required; for single-use BPA-free workflows, PETG Erlenmeyer flasks are the alternative.

How to Choose PC Erlenmeyer Flasks

Flat Base vs. Baffled Base for Your Cell Line Flat-base PC flasks produce laminar rotational flow during orbital shaking, providing low-shear mixing appropriate for shear-sensitive primary cells, some hybridoma lines, and media preparation. Baffled-base PC flasks generate turbulence during shaking that substantially increases dissolved oxygen (DO) transfer — critical for fast-growing CHO-S, HEK293-F, Expi293, bacterial (E. coli, B. subtilis), and yeast cultures where DO depletion limits cell density and productivity. NEST Scientific offers both base geometries across all standard volumes from 125 mL to 3,000 mL, with High Efficiency baffled formats extending to 5,000 mL for production-scale operations. Vented Cap vs. Plug Seal Cap Vented caps (0.22 µm hydrophobic PTFE membrane, HDPE housing) allow continuous gas exchange during shaker culture while maintaining a sterile barrier. The NEST oversized vented cap membrane provides greater gas transfer area than standard-dimension vented caps, supporting high-density cultures. Plug seal caps are gas-impermeable HDPE caps used for static media storage in PC flasks, transport between incubators, and autoclave cycles (caps must be loosened 1–1.5 turns before autoclaving to prevent pressure buildup). Never use a sealed plug cap on a flask in an orbital shaker incubator — medium acidification and DO depletion occur within hours. Volume Selection and Fill Ratio NEST Scientific PC flasks are available in 125 mL, 250 mL, 500 mL, 1,000 mL, 2,000 mL, 3,000 mL, and 5,000 mL (High Efficiency). Working fill for shaker culture: 20–25% of nominal volume. Working volumes: 25–30 mL in 125 mL; 50–60 mL in 250 mL; 100–125 mL in 500 mL; 200–250 mL in 1,000 mL; 400–500 mL in 2,000 mL. Scale-up from T-flask to Erlenmeyer: transition from T75 (15–25 mL static) to a 125 mL PC baffled flask when moving to suspension-adapted cell lines requiring orbital shaking. Autoclave Protocol for PC Flasks PC Erlenmeyer flasks can be autoclaved at 121 °C for 15–20 minutes (standard gravity cycle). Loosen cap 1–1.5 turns before autoclaving; retighten after cooling. Repeated autoclaving causes progressive hydrolytic degradation of carbonate linkages — inspect for crazing, discoloration, or dimensional distortion after each cycle and retire flask at first sign of damage. Do not use autoclaved PC flasks for vacuum applications. BPA Consideration Polycarbonate contains bisphenol A (BPA) as an inherent part of its monomer chemistry. For applications involving estrogen receptor–positive cell lines (MCF-7, T47D, ZR-75), primary endocrine cells, or long-term culture runs exceeding 14 days, PETG Erlenmeyer flasks (BPA-free) are the safer choice. For standard bacterial, yeast, CHO, and HEK293 suspension culture, trace BPA from USP Class VI PC does not affect results in published culture protocols.

What are PETG Erlenmeyer Flasks?

PETG (polyethylene terephthalate glycol) Erlenmeyer flasks are single-use, sterile, BPA-free conical shake flasks manufactured from glycol-modified PET for suspension cell culture, microbial fermentation, and media preparation on orbital shakers. PETG offers optical clarity comparable to PC, a working temperature range of −80 °C to +60 °C, superior impact resistance at low temperatures versus standard PET, and a BPA-free composition — advantages over PC in single-use workflows where autoclaving is not required, and BPA-sensitive cell culture applications are involved. PETG Erlenmeyer flasks are the correct choice for high-throughput single-use suspension culture, BPA-free applications, and labs that want to eliminate autoclave and washing steps in their shake flask workflow.

How to Choose PETG Erlenmeyer Flasks

PETG vs. PC for Shake Flask Culture PETG flasks are single-use and cannot be autoclaved (maximum temperature +60 °C). PC flasks can be autoclaved 5–10 times and have a broader temperature range (−135 °C to +135 °C). For high-throughput suspension culture operations processing many flasks per week, single-use PETG eliminates all washing, autoclave queuing, and glassware inspection steps — reducing hands-on time and contamination risk from reuse. For labs with lower throughput that prepare media in-house via autoclave, PC remains the cost-effective choice. PETG is BPA-free; PC contains BPA. Cell Line Compatibility PETG Erlenmeyer flasks are validated for CHO-S, HEK293-F, Expi293, Jurkat, K562, Sf9 (Spodoptera frugiperda), Hi5 (Trichoplusia ni), S. cerevisiae, P. pastoris, and E. coli suspension culture. The non-treated PETG surface prevents inadvertent cell adhesion and is compatible with all standard suspension culture media, including FreeStyle CHO, Expi293, Sf-900, and ESF 921 insect cell media. PETG is chemically resistant to DMSO, ethanol, isopropanol, and standard aqueous buffers. Baffled vs. Flat Base for PETG Flasks Baffled-base PETG Erlenmeyer flasks are the standard for oxygen-demanding suspension cultures: CHO, HEK293-F, bacterial, and insect cell lines. The baffles generate turbulent flow during orbital shaking, raising dissolved oxygen and reducing cell sedimentation. Flat-base PETG flasks are used for media preparation, gentle mixing, and shear-sensitive cell lines where turbulent conditions cause cell damage. NEST Scientific offers baffled and flat-base versions in both 125 mL and 1,000 mL. Vented Cap Gas Exchange NEST Scientific PETG Erlenmeyer flasks ship with a 0.22 µm hydrophobic PTFE membrane vented cap in an HDPE housing with an oversized vent area. For suspension culture with 20–25% fill volume, vented caps at standard orbital speeds (100–150 RPM) provide adequate oxygen mass transfer to support mammalian cell densities up to approximately 5 × 10⁶ cells/mL. For cultures targeting higher densities or with very high specific oxygen uptake rates (sOUR), confirm dissolved oxygen by inline monitoring before scaling further in PETG flasks. Storage of Unused Sterile Flasks Store unused NEST Scientific PETG Erlenmeyer flasks at room temperature (15–30 °C) in the original sealed sterile bag, away from UV light. Shelf life is typically 3 years from the production date per the lot certificate. Inspect the bag seal before use; a compromised seal invalidates sterility. Remove from the bag inside a biosafety cabinet immediately before inoculation.
