Bacterial and fungal RNA extraction kits isolate total RNA from organisms with rigid, recalcitrant cell walls - Gram-positive bacteria, Gram-negative bacteria, yeasts, filamentous fungi, algae, and protozoa - using bead-beating mechanical lysis combined with chaotropic guanidinium-based lysis buffers and silica-membrane spin columns or magnetic beads. Standard mammalian RNA kits cannot disrupt the thick peptidoglycan cell walls of Gram-positive bacteria or the chitin-rich walls of fungi; bead beating at 4,000-6,800 rpm using ultra-high-density BashingBeads or zirconia/silica beads is the only reliable method for complete lysis of these organisms. Use a bacterial/fungal-specific RNA kit whenever your sample contains Gram-positive bacteria, yeast, or filamentous fungi - standard total RNA kits will produce unacceptably low yields from these organisms.

How to Choose a Bacterial and Fungal RNA Extraction Kit

 

Spin-Column vs. Magnetic Bead Format

Spin-column bacterial/fungal RNA kits (Zymo Quick-RNA Fungal/Bacterial Mini/Microprep) are the standard format for 1-24 samples per run, completing extraction in minutes after bead beating.
