Prepacked flash chromatography cartridges are single-use in most workflows -- the stationary phase is contaminated by sample components and mobile phase residues after each run, and reuse degrades resolution. Empty reusable column hardware (GlasPure-style glass columns) can be repacked with fresh bulk silica multiple times, making them cost-effective for large-scale or high-frequency purifications above 5 g per run. For regulated pharmaceutical workflows, single-use prepacked cartridges are required to ensure batch-to-batch separation consistency.
Normal-phase silica cartridge loading is approximately 1 to 5% of the silica bed mass for routine separations. A solid loader is a short open-bed tube containing dry silica or Celite onto which the crude sample is adsorbed before loading onto the main flash column. It is used when the sample is not directly soluble in the initial mobile phase composition at the required concentration, or when direct liquid injection causes band broadening at the column head. The adsorbed sample distributes evenly across the stationary phase during loading, improving peak shape. Continuing to run with a degraded lamp reduces detection of low-UV-absorbing peaks and increases the risk of missed fractions.
