What are Reaction Tubes?

Reaction tubes are polypropylene (PP) single-use or low-retention microcentrifuge and PCR tubes used as the primary vessel for molecular biology reactions—PCR amplification, reverse transcription, restriction digestion, ligation, nucleic acid precipitation, protein extraction, and centrifugation of microvolume samples. Reaction tube quality directly affects PCR sensitivity, sample recovery, and contamination risk. Choose PCR-clean or PCR-grade tubes—certified free of DNA, DNase, RNase, and PCR inhibitors—for any molecular biology reaction where contamination would invalidate the result.

How to Choose Reaction Tubes

Volume and Format: 0.2 mL vs. 0.5 mL vs. 1.5/2.0 mL
0.2 mL thin-wall tubes and strip tubes are the standard PCR format; consistent wall thickness (≤0.2 mm) ensures efficient heat transfer between the thermal cycler block and the sample. 0.5 mL tubes are used for reactions generating small volumes that benefit from a snap-cap format. 1.5 mL and 2.0 mL hinged-lid tubes are the universal format for sample prep, restriction digests, ligation, protein assays, centrifugation, and short-term sample storage.

Purity Grade: Standard vs. PCR-Clean vs. Certified RNase/DNase-Free
Standard polypropylene tubes are adequate for general sample storage and centrifugation. PCR-clean tubes are certified free of DNA, DNase, RNase, and PCR inhibitors per lot testing — required for any PCR, RT-PCR, or sequencing preparation workflow. For RNA work, additionally confirm RNase-free certification. Eppendorf Safe-Lock Tubes (PCR clean grade) carry a two-layer resealable packaging system that maintains purity until the point of use.

Low-Retention vs. Standard Polypropylene
Low-retention PP is treated to reduce protein and nucleic acid adsorption to the tube wall. For reactions using <50 µL of a dilute nucleic acid or protein sample, low-retention tubes can recover 15–30% more material than standard PP by reducing wall-adsorption losses. Use low-retention tubes for single-cell, low-copy-number, or precious-sample workflows.

Hinged Lid (Safe-Lock) vs. Snap-Cap
Hinged-lid tubes (Eppendorf Safe-Lock style) have a locking latch that prevents accidental opening during centrifugation and vortexing, retaining samples at forces up to 21,300 × g. Standard snap-cap tubes may open at high g-forces, causing sample loss and cross-contamination. Choose Safe-Lock format for any step involving high-speed centrifugation or vigorous mixing.

Color Coding
Color-coded reaction tubes (natural/clear, yellow, red, blue, green) improve sample tracking at the bench and reduce the risk of mix-ups in multi-sample workflows. NEST Scientific offers colored tube collections. Confirm that the colorant does not interfere with spectrophotometric measurements (OD at 260/280 nm) if measuring nucleic acid concentration directly in the tube using a microvolume spectrophotometer.

FAQ

Reaction tubes are 0.2 mL to 2.0 mL polypropylene vessels used as the primary container for molecular biology reactions including PCR amplification, reverse transcription, restriction enzyme digestion, ligation, nucleic acid precipitation, microvolume centrifugation, and short-term sample storage. The 0.2 mL thin-wall format is the PCR standard; the 1.5 mL and 2.0 mL hinged-lid tube is the universal microcentrifuge tube for all other sample preparation steps.
PCR-clean (also called PCR grade) reaction tubes are certified free of human DNA, DNase, RNase, and PCR inhibitors per lot testing. Contamination of these components in a reaction tube produces false-positive PCR signals, RNA degradation, or inhibited amplification. Eppendorf Safe-Lock Tubes in PCR-clean grade use a two-layer resealable packaging system to maintain purity from manufacture through point of use in the laboratory.
Eppendorf Safe-Lock Tubes (1.5 mL and 2.0 mL) are rated for centrifugation at up to 21,300 × g. The locking hinge prevents accidental lid opening during high-speed centrifugation and vigorous vortexing — a critical safety feature that standard snap-cap tubes do not provide. Confirm the g-force rating for any centrifuge step exceeding 14,000 × g before using non-Safe-Lock format tubes.
Low-retention (LoBind) polypropylene tubes reduce nucleic acid adsorption to tube walls by up to 90% versus standard PP, making them the correct choice for reactions using dilute samples below 10 ng/µL DNA or 100 ng/µL protein. Applications where low-retention tubes improve recovery include single-cell sequencing library preparation, low-copy-number RT-PCR, CRISPR gRNA preparation, and any workflow where sample volume is ≤10 µL.
PCR strip tubes (8-tube or 12-tube strips) are physically connected strips of 0.2 mL thin-wall tubes that fit multi-well thermal cycler blocks and allow simultaneous capping and uncapping of all 8 or 12 tubes — reducing handling time in high-throughput PCR setups. Individual 0.2 mL tubes are used in lower-throughput workflows or when samples must be handled independently at each step. Both formats have equivalent wall thickness and heat transfer properties.
NEST Scientific 1.5 mL and 2.0 mL microcentrifuge tubes are manufactured to Eppendorf-compatible dimensions under ISO 9001:2015 quality management, providing dimensional interchangeability with Eppendorf microcentrifuge rotors and tube racks. NEST tubes offer a cost-competitive alternative for high-volume routine applications where the additional PCR-clean or LoBind certifications of premium Eppendorf grades are not required.
Colored reaction tubes are suitable for sample tracking and bench organization, but should not be used for direct spectrophotometric measurement (OD 260/280 nm) in the tube itself. The colorant absorbs UV light and produces inflated or variable OD readings. For nucleic acid quantification directly in the tube, use only clear/natural (uncolored) PP reaction tubes certified free of UV-absorbing additives.
MBP accepts PO-based bulk orders for Eppendorf and NEST Scientific reaction tubes in case quantities (500 and 1,000 packs for 1.5/2.0 mL formats; trays of 1,000 for 0.2 mL PCR formats) with direct human support for institutional procurement accounts.
