What are Round Well Deep Well Plates?

Round well deep well plates are polypropylene microplates manufactured to ANSI/SLAS (SBS) footprint standards, featuring cylindrical wells that provide greater sample volumes than standard PCR or assay plates. Typical formats include 96-well plates with capacities of 1.2 mL, 2.0 mL, or 2.2 mL per well, designed for automated sample processing, nucleic acid extraction, microbial culture, compound storage, and high-throughput laboratory workflows.

The round well geometry promotes uniform mixing, consistent liquid movement during shaking, and efficient magnetic bead collection during automated extraction procedures. Unlike square well plates, which maximize storage volume, round well plates are preferred when sample recovery, mixing performance, and magnetic bead handling are critical.

How to Choose Round Well Deep Well Plates

Well Volume

Select well volume based on sample input and workflow requirements.

  • 1.2 mL plates are ideal for lower-volume nucleic acid extraction, sample preparation, and compact storage.
  • 2.0 mL plates are the most commonly used format for DNA, RNA, and protein extraction workflows.
  • 2.2 mL plates provide additional working capacity for larger sample volumes, bacterial culture, and high-yield extraction protocols.

Always verify both total well volume and recommended working volume before use.

Bottom Geometry

The well bottom design affects sample recovery and mixing performance.

  • U-bottom wells support efficient mixing and magnetic bead collection, making them the standard choice for extraction workflows.
  • V-bottom wells concentrate liquids and pellets at a single point, improving aspiration efficiency and pellet recovery.
  • Flat-bottom wells are occasionally used when optical measurements or specialized assay workflows require a flat surface.

Sterility Requirements

Non-sterile plates are suitable for most nucleic acid extraction, sample storage, and analytical workflows.

Sterile, gamma-irradiated plates (SAL 10⁻⁶) are recommended for:

  • Cell culture applications
  • Microbiology workflows
  • Live-cell processing
  • RNA extraction from cultured cells

Centrifugation and Sealing

Most polypropylene round well plates withstand centrifugation between 4,000 × g and 6,000 × g, depending on design.

For storage and automated processing, confirm compatibility with:

  • Adhesive sealing films
  • Heat-seal foils
  • Silicone sealing mats
  • Automated plate sealers

Raised well rims improve sealing performance and reduce evaporation during storage.

Automation Compatibility

Quality round well deep well plates conform to ANSI/SLAS dimensions (127.76 × 85.48 mm), ensuring compatibility with automated platforms including:

  • Hamilton liquid handlers
  • Tecan systems
  • Beckman Coulter platforms
  • Agilent automation systems
  • Magnetic bead processors

Alphanumeric indexing (A–H and 1–12) supports automated sample tracking and workflow management.

Specifications Context

Round well deep well plates are manufactured from high-purity polypropylene and are commonly rated for temperatures from −80°C to +121°C, supporting freezer storage and autoclave sterilization. Premium plates are produced from USP Class VI resins and tested for DNase, RNase, DNA, and PCR inhibitor contamination.

The 96-well 2.0 mL U-bottom round well plate remains one of the most widely used formats for automated nucleic acid extraction, biobanking, bacterial culture, and sample preparation workflows due to its balance of volume capacity, automation compatibility, and reliable magnetic bead performance.

FAQ

Round well deep well plates are 96-well polypropylene microplates with cylindrical or conical round-bore wells, typically available in 1.2 mL, 2.0 mL, and 2.2 mL per well. They are preferred over square well plates for magnetic bead-based extraction workflows — round well geometry provides better bead collection against the well wall during magnetic separation — and for protocols requiring orbital mixing or spinning where liquid meniscus behaviour in round wells is more predictable.
Round well deep well plates are most commonly available in 1.2 mL, 2.0 mL, and 2.2 mL per well in 96-well format. The 1.2 mL format is specified for smaller sample input volumes and lower deck height requirements on automated platforms. The 2.0 mL round well format is standard for KingFisher Flex nucleic acid extraction workflows with 100–1,000 µL input samples.
Round well deep well plates are used as staging vessels in some KingFisher protocols, but the KingFisher Flex specifically requires the Thermo Scientific KingFisher-branded deep well plate (Cat. No. 95040450) whose well geometry and depth are precisely matched to the magnetic rod travel path. Standard round well polypropylene deep well plates from other manufacturers can serve as reagent staging plates but should not substitute for KingFisher-specific plates in the bead-processing positions without protocol validation.
Round well deep well plates are available with U-bottom (hemispherical, for mixing and culture), V-bottom (conical, for complete aspiration and pellet concentration), and flat-bottom (for culture and assay applications requiring an optical flat surface) configurations. U-bottom round wells are the most common for general sample storage and nucleic acid extraction; V-bottom round wells concentrate pellets for centrifugation and are preferred in enzymatic assay and precipitation protocols.
Virgin polypropylene round well deep well plates are autoclavable at 121 °C / 15 PSI for standard liquid-cycle sterilization, making them suitable for sterile bacterial culture and cell-based assay workflows at the 1.2–2.0 mL per well scale. Sterile gamma-irradiated options (SAL 10⁻⁶) are also available for workflows where autoclave access is not practical or where pre-sterilized single-use plates are preferred.
Round well deep well plates conforming to the ANSI/SLAS 1-2004 standard have a footprint of 127.76 mm (length) × 85.48 mm (width), with ±0.5 mm tolerance. This footprint is universal across Hamilton, Tecan, Beckman, and Agilent liquid handling platforms and standard centrifuge buckets. Plates not meeting this standard may misalign on automated decks and fail flatness testing requirements.
For nucleic acid extraction and molecular biology, specify round well deep well plates certified DNase-free, RNase-free, DNA-free, and free from PCR inhibitors, manufactured from virgin polypropylene without mold release agents. Lot-specific CoA documentation is required for validated clinical or regulated workflows. USP Class VI polypropylene resin certification is available on select lines for GMP-adjacent applications.
