TC-treated (tissue culture–treated) cell culture plates are sterile, single-use multi-well polystyrene plates whose well surfaces have been modified by vacuum plasma treatment to introduce negatively charged oxygen-containing groups (carboxyl, carbonyl, hydroxyl), reducing the water contact angle from ~75–90° (untreated PS) to below 10° (hydrophilic). This surface modification directly enables attachment, spreading, and monolayer formation of adherent mammalian cell lines. TC-treated plates are the default format for all adherent mammalian cultures, cell-based assays, ELISA preparation, compound cytotoxicity screening, transfection, and gene expression studies.

Well Format by Cell Number and Assay Type
6-well TC-treated plates (9.4 cm²/well) are the standard for Western blot preparation, siRNA transfection, viral infection, protein overexpression, and colony-forming unit (CFU) assays requiring ≥3 × 10⁶ cells per condition. 12-well plates (3.8 cm²/well) suit medium-scale reporter assays, luciferase quantification, and ELISA from cell lysate. 24-well plates (1.9 cm²/well) are versatile for cytotoxicity dose-response in 4–6 conditions per plate, primary cell experiments, and viral plaque assays. 96-well flat-bottom TC-treated plates (0.32 cm²/well) are the universal format for cell viability (MTS, MTT, CCK-8), compound screening, ELISA, reporter gene assays, and qPCR lysate preparation across 8–12 concentrations with triplicate wells. 384-well plates reduce reagent cost per compound for HTS drug discovery programs.
Lot-to-Lot TC Treatment Consistency
For longitudinal drug screening, biomarker validation, or multi-site studies, lot-to-lot variation in TC treatment can alter cell attachment density, proliferation rate, and assay Z-factor. NEST Scientific verifies vacuum plasma treatment by water contact angle goniometry on each manufacturing lot; the release criterion is contact angle <10°. Low-Evaporation EDGE Plates
For 96-well dose-response assays running 72–96 hours, peripheral (edge) well evaporation can increase medium osmolality in the outer ring by 10–20% versus inner wells, introducing a systematic bias in IC50 determinations. NEST Low Evaporation EDGE 96-well TC-treated plates use an internal well ridge design that reduces edge-well evaporation during extended incubations. Use EDGE plates for any dose-response study where incubation time exceeds 48 hours.

Do NOT use TC-treated plates for Jurkat, K562, HL-60, suspension CHO-S, hybridomas, Sf9, Hi5, or any non-adherent cell line.
