Vacuum Filtration

What are Vacuum Bottle-Top Filter Systems?

Vacuum bottle-top filter systems are single-use, sterile filtration assemblies consisting of a graduated polystyrene funnel with an integrated membrane filter bonded to a polystyrene receiver bottle, connected by a hose-port adaptor to a house vacuum line or benchtop vacuum pump. The applied negative pressure (−50 to −100 kPa) draws liquid through the membrane into the receiver bottle, which is sealed after filtration for immediate use or cold-room storage.

How to Choose Vacuum Filtration Systems

PES vs. PVDF Membrane for Vacuum Filtration PES membranes provide low protein binding (~1–2 µg/cm²), high flow rates, and broad aqueous-solution compatibility — the standard for routine DMEM, RPMI, HBSS, PBS, and supplement-containing media sterilization. PVDF membranes have slightly lower protein binding than PES and greater chemical resistance to mild organic solvents, mild acids, and alkaline solutions. Use PVDF when filtering protein-rich solutions (FBS, antibody preparations, conditioned medium with high protein content) or when mild organic solvents are present in the filter solution. Volume Selection 250 mL systems (50 mm diameter membrane, 50 mL funnel) suit small media aliquots, supplement solutions, and transfection reagent preparations. 500 mL systems (90 mm diameter membrane, 150 mL funnel) are the routine lab workhorse for standard 500 mL media bottle preparation and FBS filtration. 1,000 mL systems (90 mm diameter membrane, 350 mL funnel) handle full-liter media preparations and large-volume buffer sterilization. All NEST systems include a pre-attached, graduated polystyrene receiver bottle in the corresponding volume. Pore Size: 0.10 µm vs. 0.22 µm vs. 0.45 µm 0.22 µm is the sterilizing grade per USP <1229.2> and ICH Q9: retains all bacteria and fungi; used for routine cell culture media, PBS, and buffer preparation. 0.45 µm is the clarification grade: used as a pre-filtration step before 0.22 µm to remove large particulates from turbid or high-biomass samples (FBS, conditioned media, cell culture supernatants, crude lysates), extending 0.22 µm filter throughput by 3–5× on difficult samples. 0.10 µm is the mycoplasma-removal grade: required when preparing media for Mycoplasma-sensitive primary cell culture or when processing serum or supplementary reagents that may harbor Mycoplasma species. Note that 0.10 µm filtration is substantially slower than 0.22 µm due to the tighter membrane pore structure. Practical Tips for Maximum Throughput Pre-wet the membrane by briefly passing 5–10 mL of sterile water through before the sample — this wets the hydrophilic PES or PVDF membrane and reduces initial resistance. For FBS and other viscous solutions, pre-filter through 0.45 µm before the 0.22 µm step. Maintain a vacuum at −50 to −80 kPa; excessive vacuum (> −100 kPa) can shear fragile membrane pores and increase non-specific protein adsorption. Do not autoclave the assembled bottle-top filter — PES and PS components are not rated for autoclave sterilization.

Specifications Context

Cell Scrapers

What are Cell Scrapers?

Cell scrapers are sterile, single-use disposable tools with a polystyrene handle and a free-rotating polyethylene blade used to mechanically detach and harvest adherent mammalian cells from the TC-treated surface of culture flasks, dishes, and multi-well plates when enzymatic detachment (trypsin, Accutase, Dispase) is not appropriate. The free-rotating blade mechanism allows the angle to be adjusted by applying gentle pressure on the handle — the blade twists to align with the vessel surface for consistent, complete cell removal across the full growth area. Choose the 13 mm blade for T25 and T75 flasks and multi-well plates; choose the 20 mm blade for T175, T225, and 100–150 mm dishes, where a wider blade reduces the number of passes needed to clear the full growth surface.

How to Choose Cell Scrapers

When to Use Cell Scrapers Instead of Trypsin Cell scraping is the correct harvesting method when:

Trypsinization damages the protein or surface marker of interest; scraping preserves surface antigens, including CD markers for flow cytometry and receptor proteins for biochemical assays.

The cell line does not detach effectively with trypsin (some primary cells, fibroblasts in dense monolayers, epithelial cells after extended culture)

Harvest must be complete and rapid for protein or RNA extraction protocols with RNase-sensitive targets, where trypsin incubation time introduces RNA degradation risk

The downstream assay uses cells in medium without trypsin carryover (some enzyme assays, reporter gene assays, and cell viability assays where trypsin activity in the sample affects results).

Blade Size by Vessel Format The 13 mm blade (catalog 710001, 220 mm handle) is designed for T25 and T75 flasks and multi-well plates (6-well through 24-well). The blade width covers approximately 1–1.5 passes across a T75 flask width for complete surface coverage. The 20 mm blade (catalog 710011, 280 mm handle) suits T175 and T225 flasks, 100 mm and 150 mm dishes, and any large-surface vessel where the wider blade reduces scraping passes needed and operator fatigue. Both blade sizes fit through the standard T-flask neck opening. Free-Rotating Blade Mechanism NEST Scientific cell scrapers use a free-rotating blade that pivots relative to the handle on a hinge mechanism. Applying gentle lateral pressure on the handle adjusts the blade angle, allowing it to maintain flush contact with the vessel floor during the scraping stroke — improving cell detachment yield compared to fixed-blade scrapers, where the blade angle may not match the vessel floor geometry. After the push stroke, rotating the handle slightly returns the blade to the correct alignment for the next stroke. Scraping Technique Add 1–2 mL of culture medium or PBS to the vessel before scraping to lubricate the surface and prevent cells from drying. Work systematically across the flask or dish in parallel overlapping strokes. For T-flasks, tilt the flask at 20–30° to pool the cell suspension at the low end after scraping, then pipette out the suspension. Do not press excessively — scraping force sufficient to detach cells should not require grinding the blade into the vessel floor, which damages cells and the scraper.

Specifications Context

Cell Strainers

What are Cell Strainers?

Cell strainers are sterile, single-use nylon mesh filter caps that sit in the opening of a 50 mL conical centrifuge tube, used to disaggregate tissue homogenates, tumor biopsies, lymph nodes, spleens, and bone marrow aspirates into uniform single-cell suspensions for downstream applications including flow cytometry, primary cell culture, FACS sorting, and immunophenotyping. The strainer is placed filter-side down in the collection tube, the tissue suspension is added to the strainer basket and pushed through the nylon mesh using the plunger of a syringe, the wet end of a syringe barrel, or the back of a sterile 1 mL syringe. Choose 40 µm for fine single-cell suspension preparation from soft tissues; 70 µm for standard tissue disaggregation (the most commonly used pore size); 100 µm for pre-filtration of coarse tissue homogenates and large spheroids.

How to Choose Cell Strainers

Pore Size by Application 40 µm strainers (blue) produce the finest filtrate, retaining cell aggregates and larger tissue debris while passing single cells and small cell clusters. They are the standard for preparing highly uniform single-cell suspensions from soft tissues (spleen, thymus, lymph nodes) for flow cytometry, where doublets must be minimized. 40 µm strainers have lower flow rates and clog more readily with dense or fibrous tissues. 70 µm strainers (white) are the most commonly used pore size in cell biology — they balance efficient single-cell passage with adequate retention of fibrous connective tissue and undigested tissue chunks. Standard for tumor dissociation, liver perfusion single-cell prep, and any tissue where a moderate doublet rate is acceptable. 100 µm strainers (yellow) pass cell clusters and small organoids while retaining large undissociated tissue pieces; used as a first-pass pre-filtration step before 70 µm or 40 µm filtration of tough fibrous tissues, and for filtering 3D tumor spheroids grown in 96-well U-bottom plates before reseeding. Color-Coding for Laboratory Workflow NEST Scientific cell strainers follow a universal color-coding convention: 40 µm = blue, 70 µm = white, 100 µm = yellow. This matches the color convention used by BD Biosciences Falcon cell strainers, allowing unambiguous pore-size identification during multi-step tissue disaggregation protocols without reading the label at every step. 50 mL Tube Compatibility NEST Scientific cell strainers are designed with a flange diameter that fits securely into the opening of 50 mL conical centrifuge tubes from most major brands, including BD Falcon, TPP, and MidSci, per NEST Scientific product documentation. A molded tab on the upper ring facilitates sterile handling during tube loading without touching the mesh surface. Tissue Types and Recommended Pore Size For spleen, thymus, and lymph nodes: 40 µm or 70 µm. For bone marrow: 70 µm. For tumor biopsy (mechanically or enzymatically dissociated): 40 µm (after enzyme digestion) or 70 µm (mechanical disaggregation only). For liver (perfusion-dissociated hepatocytes): 70 µm or 100 µm. For 3D spheroid disaggregation: 40 µm or 70 µm, depending on the desired cluster size in the filtrate. For flow cytometry sorting (minimize doublets): 40 µm immediately before loading the sorter.

Syringe Filters

What are Syringe Filters?

Syringe filters are self-contained, disposable filtration units comprising a polypropylene (PP) housing enclosing a flat-sheet membrane, with a female Luer lock inlet and a male Luer lock outlet. They are attached to a standard syringe, the sample is drawn up, and pushed through the membrane by manual plunger pressure. Standard disc diameters are 4 mm, 13 mm, 25 mm, and 33 mm; membrane pore sizes are 0.22 µm (sterilizing grade) and 0.45 µm (clarification grade), with 0.10 µm available for mycoplasma removal. Effective filtration areas: 13 mm filter ≈ 0.92 cm²; 25 mm filter ≈ 2.98 cm². Globe Scientific syringe filters, a confirmed MBP brand, are available in PP housing with PES, PVDF, CA (cellulose acetate), MCE, and nylon membranes in 13 mm and 30 mm diameters, sterile (individually wrapped) and non-sterile (bulk), 0.22 µm and 0.45 µm. Choose syringe filters for volumes of 1–150 mL; choose vacuum bottle-top filters for volumes above 150 mL.

How to Choose Syringe Filters

Diameter Selection by Sample Volume 13 mm syringe filters (effective area 0.92 cm²) handle up to 10–15 mL per filter before flow resistance becomes impractical. Use with 1–10 mL syringes. 25 mm syringe filters (2.98 cm²) handle 10–100 mL per filter. Use with 10–60 mL syringes. 33 mm syringe filters (approximately 5–6 cm²) handle up to 150 mL and are the largest practical syringe filter format. For volumes above 150 mL, switch to a vacuum bottle-top filter system. Matching filter diameter to sample volume prevents premature membrane clogging and excessive plunger force. Membrane Chemistry for Syringe Filters PES (Polyethersulfone): Low protein binding, high flow rate, aqueous compatibility. Standard choice for biological sample sterilization, cell culture supplement filtration (antibiotics, glutamine, growth factors), and conditioned medium aliquots. Not suitable for strong organic solvents or concentrated acids. PVDF (Polyvinylidene fluoride, hydrophilic): Low protein binding — among the lowest of all syringe filter membranes, with protein recovery rates >90–99.9% documented for antibody and enzyme solutions. Compatible with mild organic solvents, weak acids, and most aqueous solutions. The first choice when filtering precious protein solutions (antibodies, enzymes, viral preparations) is where maximum protein recovery is critical. Not suitable for pure DMSO (use PTFE). Nylon (Polyamide): Low extractables, broad chemical compatibility including mixed aqueous-organic solvents, methanol, acetonitrile, and DMSO-water mixtures. Standard for HPLC mobile phase filtration and dissolution testing. Higher protein binding than PES or PVDF — avoid for biological samples where protein loss is a concern. MCE (Mixed Cellulose Ester): Low cost, aqueous applications only; moderate protein binding. Suitable for general buffer preparation, water sterilization, and non-critical sample preparation. Not compatible with organic solvents or strong acids. CA (Cellulose Acetate): Low protein binding, similar to PES. Suited for biological sample filtration and sterility testing applications. Contains surfactant treatment for wettability — check for extractable compatibility in downstream assays. PTFE (Polytetrafluoroethylene, hydrophilic): Maximum chemical resistance — compatible with pure DMSO, concentrated acids and bases, and aggressive organic solvents. The only membrane choice for 100% organic solvent filtration. For aqueous solutions, use hydrophilic PTFE (pre-wetted with methanol or IPA before aqueous filtration unless the hydrophilic version is selected). Sterile vs. Non-Sterile Syringe Filters Sterile syringe filters (individually gamma-irradiated or E-beam sterilized, individually packaged) are required for cell culture media sterilization, supplement preparation, and sample preparation for biological culture. Non-sterile bulk-packed filters (Globe Scientific non-sterile formats) are appropriate for HPLC mobile phase preparation, analytical sample clarification, and non-biological applications where sterility of the filtrate is not required.

Specifications Context

Globe Scientific syringe filters are manufactured in a PP housing with a luer-lock inlet and outlet conforming to ISO 594 / ISO 80369 for Luer fittings. PES and PVDF sterile filters are individually gamma irradiated; each is separately wrapped and printed with an expiry date per Globe Scientific lot protocol. Effective filtration areas: 13 mm filter ≈ 0.92 cm²; 30 mm filter ≈ 4.91 cm² (Globe Scientific welded format). Operating temperature: up to 60 °C. Do not exceed the maximum operating pressure of 6 bar (87 psi). Globe Scientific is ISO 9001:2015 certified.
