What Are AAV Control & Reporter Constructs?

Establishing reliable experimental controls is critical in AAV research because transduction efficiency varies substantially by serotype, cell type, multiplicity of infection (MOI), and batch-to-batch variation in viral preparation quality. Matched AAV control and reporter constructs provide the reference point for interpreting experimental results.

How to Choose AAV Control & Reporter Constructs

Serotype and Backbone Matching The control construct should use the same AAV serotype, ITR configuration, and promoter as the experimental construct, ensuring transduction efficiency differences reflect the transgene rather than vector or serotype variation. Fluorescent Reporter Choice GFP and mCherry reporters allow confirmation of transduction by fluorescence microscopy or flow cytometry, with mCherry offering a red-channel option for multiplexed experiments. Luciferase and Dual-Reporter Options Firefly luciferase provides quantitative bioluminescent transduction confirmation; dual reporter systems (e.g., fluorescent + luciferase) allow both visual and quantitative readouts from the same construct. Empty Vector Baseline An empty AAV vector — same ITRs, serotype, and promoter, no transgene — establishes the baseline effect of AAV capsid delivery and innate immune activation, independent of the gene of interest. Batch-to-Batch Variation Controls Because AAV preparation quality can vary between batches, including a transduction-efficiency control (e.g., GFP reporter) alongside each new batch of experimental AAV helps confirm comparable functional titer.

Specifications Context

AAV transduction can activate innate immune sensing pathways and, at high vector doses, induce cellular stress responses independently of the transgene. A matched empty vector or reporter control sharing the same serotype and promoter as the experimental construct helps distinguish vector-related effects from genuine transgene-specific phenotypes. Because AAV infectivity can vary between preparations, reporter-based transduction assessments are commonly used to verify functional performance before experimental studies. For in vivo applications, pilot reporter studies can help confirm tissue targeting and delivery efficiency before larger cohorts are committed. Product availability reflects MBP's catalogue as of mid-2026.
