On blood draw, the lysis reagent immediately inactivates nucleases and preserves both genomic DNA and RNA—including transcriptomic expression profiles—from 3–6 mL of mammalian whole blood. They are used in gene expression analysis, next-generation sequencing, PCR-based molecular diagnostics, cell-free DNA (cfDNA) studies, and biobank archiving. DNA vs RNA vs Dual Preservation
K2 EDTA tubes (lavender cap) prevent clotting and preserve cellular morphology for CBC and flow cytometry, but RNA degrades rapidly without additional stabilization. PAXgene Blood RNA Tubes (Qiagen) stabilize intracellular RNA expression profiles. Zymo Research Blood Collection Tubes stabilize both DNA and RNA simultaneously in a single draw tube, enabling dual downstream analysis from one specimen.
Blood Volume Per Tube
Standard blood collection tubes hold 3 mL or 6 mL. Confirm draw volume against your extraction kit's minimum input requirement. Many NGS and RT-PCR extraction kits require 1–3 mL of whole blood per reaction, making 3 mL tubes sufficient for a single downstream application.
Processing Window
For immediate processing (< 2 hours post-draw): K2 EDTA tubes are adequate. For delayed processing (2–24 hours at RT): nucleic acid-stabilizing tubes required. For samples needing shipping before extraction: tubes with DNA/RNA Shield-based chemistry stabilize at ambient temperature for up to 28 days without freezing.
Anticoagulant Type
K2 EDTA (dipotassium EDTA) is the preferred anticoagulant for molecular applications, including PCR and NGS. K3 EDTA (tripotassium) is a liquid formulation, sometimes preferred for automated analyzers. Citrate tubes are not recommended for nucleic acid applications. Heparin tubes are a known PCR inhibitor and should be avoided for molecular workflows.
Regulatory Compliance
For clinical or regulatory-submission workflows, confirm tube CE marking or FDA 510(k) clearance status with the manufacturer. Research-use-only (RUO) blood collection tubes are not cleared for in vitro diagnostic use.
Standard evacuated blood collection tubes use medical-grade PET or glass construction. K2 EDTA tubes require 8–10 inversions immediately post-draw to ensure complete anticoagulant mixing. DNA/RNA Shield blood collection tubes lyse cells on contact with the reagent; the sample can be processed immediately or stored at ambient temperature (15–25 °C) for up to 28 days per Zymo validation data. Nucleic acid yields from 3 mL whole blood: typically 3–15 µg genomic DNA and 1–5 µg total RNA, depending on donor and processing kit. As of 2026, Zymo Research blood collection tubes are available in configurations for both immediate processing and field-stable transport.
